It will absolutely stop flow going the wrong way but its worth noting that it has a fairly high 'cracking' pressure - I haven't got the equipment to measure it but hopefully it'll still be useful if I say you'd have to blow fairly hard to get the air moving. A light bit of pressure won't do anything.From the photo, you should be able to see 1 of 2 arrows that show the direction of flow, but as you'll appreciate the lighting has to be just right to see them.

I wanted to use this inline in a small guttering job on a rabbit hutch. The volume of water in the 6mm ID tubing wasn't enough to open the valve so was useless for my needs. However, worked in testing with higher flow rate and seems well made.
